Riteish was the first actor to make his debut in a Hindi film debut in the 2003 romance Tujhe Meri Kasam in the role of Genelia D’Souza. His first commercially successful film came in the form of Kyaa Kool Hai Hum a sexually-charged film.

He has been featured in numerous commercially popular films, including Heyy Babyy, Dhamaal, Kya Kool Hai Hum, Malamaal Weekly, Housefull, Blu Master!, Housefull 2, Ek Villain, Tere Naal Love Ho Gaya, and The Grand Masti as well as Housefull 3

Deshmukh established his own production company Mumbai Film Company in 2013 by directing the Marathi film Balak Palak which received numerous awards. The year 2013 saw him create his own cricket squad Veer Marathi in the Celebrity Cricket League which he was the owner of and mentored. He was captain of the team, owned by Dheeraj Deshmukh, his brother. Dheeraj Deshmukh, while the brand ambassador is his wife Genelia D’Souza.
